Objectives: To investigate and select a number of factors for the formulation of liposomal amphotericin B (AmB) preparation by film hydration method. Materials and methods: Liposomal AmB is prepared by film hydration method with main ingredients including: AmB, hydrogennated
soy phosphatidylcholine (HSPC), disteroyl phosphatidylglycerol (DSPG), cholesterol (Chol). Survey and select those factors: The ratio (%) of mol AmB/total phospholipid (PL); and molar HSPC/DSPG; types of hydration solution based on the criteria of organoleptic properties, particle size, particle size distribution (PDI) and the percentage of entrapment of AmB. Results: The ratio of mol AmB/total amount of PL was 15%; mol HSPC/DSPG ratio was 2.0:0.8; the hydration solution was the citrate solution pH 5.0 for the formula of liposomal AmB preparation by film hydration method.
